Post-Doctoral position at the ENS

A 2-3 years post-doctoral position is available at the ENS, Paris, to study the initiation and growth of cancer from a single cell in zebrafish. The idea is to use the optogenetic tools developed in the host lab to turn on one (or a few) oncogenes (or off some tumor suppressor genes) in single cells of a zebrafish larvea. The probability of tumorigenesis will be measured and the genetic analysis of cancer developing cells vs non-cancer developing ones will be conducted.

For this project we are looking for a smart, independent and driven person with demonstrated experience in zebrafish: micro-injection, transgenesis, microscopic observations, ISH and genetic analyses.
Knowledge of image analysis (ImageJ) and computer programming (R, Matlab, LabView) favored.

The person will be paid a net post-doctoral salary of about 2800€ (depending on experience).

Au sein de l’équipe R&D Microfluidique, vous avez la responsabilité complète de l’optimisation des voies de synthèse et de l’amélioration des propriétés émulsifiantes de polymères fluorés. Vous assurez la caractérisation physico-chimique des molécules et évaluez la performance de vos nouveaux produits avec l’objectif de les produire à l’échelle requise pour la commercialisation.

Vos missions :
- Comprendre les mécanismes de synthèse existant et leurs limites
- Concevoir et synthétiser des polymères fluorés innovants
- Evaluer les performances des émulsions créées à partir de ces nouvelles molécules
- Simplifier et rendre plus robustes les voies de synthèses existantes et transmettre les améliorations à l’équipe en charge de la production
- Améliorer les protocoles d’analyses en place pour les rendre plus simples et plus rapides
- Développer de nouvelles méthodes pour des applications spécifiques (récupération de gouttelettes, diminution du volume mort,…)
- Interagir avec les différents services pour imaginer les futurs produits et concevoir les prototypes

Vos atouts :
- Doctorat en physico-chimie ou biochimie, idéalement appliqué aux microfluides, sciences des matériaux ou équivalent
- Une expérience complémentaire confirmée en chimie des polymères fluorés (post-doc,…)
- Une première expérience en microfluidique des gouttelettes

Formation et compétences :

Depuis 30 ans, le Service de Pharmacologie et d’Immunoanalyse (SPI) du CEA/Saclay démontre son expertise dans le développement de méthodes immunoanalytiques, d’analyses physicochimiques (LC-MS en particulier) et de culture cellulaire lui permettant d’aborder de multiples questions concernant la détection, la quantification et le devenir de molécules de natures diverses et variées.

Dans le cadre d’un programme de recherche du Laboratoire d’Etude du Métabolisme des Médicaments (SPI/LEMM), il s’agira de développer de nouvelles approches de spectrométrie de masse et de protéomique ciblée pour la caractérisation et la quantification absolue de toxines protéiques au sein de matrices complexes. Ces outils analytiques permettront de répondre à des besoins pour le diagnostic et la surveillance en santé publique. De bonnes capacités rédactionnelles et de communication sont demandées.

Profil : Chimiste (analytique) maitrisant les techniques de spectrométrie de masse (LC-MS/MS, Maldi-TOF) pour la quantification et la caractérisation de protéines, et de biochimie (extraction, digestion de protéines, etc…).

The international SIREDO Oncology Center (Care, Innovation, and Research in pediatric, adolescent and young adult oncology) was created in 2017 and is headed by Dr. Olivier Delattre. The center aims to improve patient’s care through extensive comprehensive studies of the biology of the young patient’s tumors. It includes Researchers, Physicians and Health care professionals with the aim of facilitating interactions between researchers and doctors, from bench to bedside and reciprocally. Our main strength is our capacities to start from fundamental research, aimed at better understanding the biology and development of the tumors, including Neuroblastoma, Rhabdoïd tumors, sarcomas, medulloblastoma, retinoblastoma to preclinical and clinical studies.

PROGRAM OVERVIEW
We aim to recruit highly talented young scientists from all over the world who wish to pursue research projects in basic and translational pediatric cancer research. The postdoctoral fellow can join one of the following research teams at SIREDO:

A full-time postdoctoral position is available in the « Molecular Regulation of Cancer Immunity » team of the Cancer Research Center of Lyon, France. The team studies the NF-kappaB family of transcription factors in T-cell biology, with a focus on tumor immunity.

Research project
Understanding the molecular mechanisms orchestrating adaptive immune responses to cancer is of the highest interest to identify novel therapeutic strategies. We have recently shown that discrete NF-kB subunits display selective functions in the biology of Foxp3+ regulatory T cels (Treg) in cancer and autoimmunity. Intriguingly, our preliminary data suggests that NF-kB may also be implicated in regulating the biology of Teff cells. The current project therefore aims at understanding the roles of the different NF-kB subunits in Teff cells in the steady-state and in the context of melanoma. The candidate will utilize unique mouse models, patient samples and high-throughput sequencing to evaluate the contribution of NF-kB to Teff cell biology.

Scientific environment
The CRCL provides resources to conduct cutting edge collaborative research, outstanding intellectual environments and state-of-the-art facilities. The team was allocated a Chair of Excellence fund of the Labex DEVweCAN that guarantees high level scientific research. The team is currently composed of 4 members working collaboratively and seeks for rapid expansion. The candidate will present her/his work at the weekly lab meeting and monthly seminars of the Immunity, Virus and Inflammation Department. The candidate will be supervised by the PI (YGB) and is expected to train students. The lab language is English.

Applicant profile
The candidate must hold a Ph.D. (or be about to graduate) in immunology. Experience in flow cytometry and animal handling is mandatory. Working knowledge in cancer biology, biochemistry or bioinformatics is a bonus. We are looking for a hard-working, highly motivated candidate, who will be actively looking for funding opportunities and will be involved in writing grants and research papers.
